Is there any way to do this in the plugin file before I upload to wp? I have a lot of sites and don't want to do it on all sites, rather do it once and upload the nulled plugin to all sites.

there is no way
you must install manually for each site you have

Update : it's not updating for me. I even try deleting and re-installed the newest, still on the previous version. Is it because I don't have WPMU dev dashboard installed ??
